Hi! Gram stainaing is not used on eukaryotes because it stains the peptidoglycan layer of the bacterial cell wall. The gram (+) or Gram (-) bacteria have differences in the thickness of this layer which differentiates the two general classes o0f bacteria from each other. The eukaryotic cell membrane has no peptidoglycan layer.
