Can you drive ? if you are coming from the states you can drive through central America and then through Colombia, if the guerrilas dont get you. There there is an old military road through the Amazon. It passes over wooden bridges and some of the panels in the bridges are missing, but you can still make it if you are very careful, dont drive in the rainy season as you will get suck in the mud. You need plenty of food and water, spaer parts for your cars the nearest repair station could be hundreds of miles away. Watch out for Alligators, Pirana fish, Cobras, Anacondas, and oh suspicious native Indians whom live deep in the Amazon. Apart from that the journey will be never to be repeated experience for you.
