The only way that you can blow a fuse unhooking a water heater is if you do not remove the fuses that protect that circuit from the fuse box when doing so. A water heater will have two fuses protecting the circuit. If you only remove one fuse one of the wires to the unit is still live and can short to ground or zap you really good which could cause injury or death.

Yes, if you didn't shut it down the power first.
You say "fuse", so you probably have cartridge type fuses in a carrier for the water heater. These are usually at the top of the panel, and have a handle on them...you pull them out to disconnect the power or replace the fuse. You might have two round plug type fuses, one for each phase of the 220V power. If you don't unscrew both, you can get a nasty, potentially fatal, surprise. What I do is when I'm sure I've shut down the power, and my meter says it's dead...before I grab any wire I tap them with the back of one hand, with fingers towards me. Often, if you grab a hot wire, your muscles clench up...causing you to hold on even tighter! If you do a final check with the back of your hand like that, if it's still hot, your hand and arm will clench up, but away from the wiring.

Yes you can: Unhook the breaker first, then check it to see that it is indeed off, then proceed to check the wires at the heater, if everything if off then proceed to unhook the heater.

yes, it is possible, but if you do it should be easy to fix, just turn the circut breaker back on, or replace the fuse

yes in fact if the power is on your most likely GOING to trip a breaker and possibly hurt yourself
