There are a lot of diseases in the US.
Or

"Many kinds of diseases" would be the best. I'm good at english. Diseases can be counted so many is ok. Lot is for things that are uncountable- a lot of water, a lot of mud...

If you have to answer from one of those listed, I would go with #2, lot of kinds of diseases. that is the most grammatically correct given the choices.

"lots of kinds of diseases" or "a lot of kinds of diseases" seem appropriate, but sound a little odd. I think saying "many kinds of diseases" sounds more correct. But maybe that's just me...? :)

Many diseases
Several diseases
Many kinds of diseases
Several kinds of diseases
