Interesting question. I'm really not sure who'd play the main characters. The only "little girl" actor I can think of is Dakota Fanning but she'd be too old to play JonBenet. Really I don't know enough about John and Patsy to know who'd play them..

For John Mark Karr, I think Martin Short could portray him well. As for Jon Benet, I bet they'd get Dakota Fanning since she gets cast for every movie that needs a young girl. As for her parents and brother, I'm not sure who'd they cast.
