If you just want to take your computer off the network, then both of the above suggestions will work.
If you mean you want to remove a certain IP address from bieng used, you'll have to put it in as "Reserved" on the DHCP server and don't assign it to any MAC address.

Run the Command Prompt from Start, Run... and type in cmd and press enter. Or from All Programs, Accessories, and click on Command Prompt.
Type in this single command "ipconfig/release", this will release the IP address for the specified adapter. Thus your IP address will be all set to 0.0.0.0 hence disabling network connection to your PC. You can view your IP address by typing "ipconfig".
To enable back your IP address, type "ipconfig/renew" and you should be able to connect to the network.

You cant disable your IP, but you can disable your LAN connection ( your NIC card). Go into Network Places, right click on your connection, and choose Disable.
