I think it has to be a tie between Richard Gere & Julia Roberts, (Pretty Woman and Runaway Bride) and Keanu Reevees & Sandra Bullock, (Speed and The Lake House). The on screen chimisry between these talented and gifted actors/acctresses is so electric that the audience feels it and can't help but react to it. You leave knowing you gotten more than your money's worth.

Humphrey Bogart & Lauren Bacall, in several movies. Its very obvious they had the major hots for each other.
Best male chemistry is Jack Lemmon & Walter Matthau in "The Odd Couple"..just brilliant double act.

Frodo and Samweis, you just can't beat that chemistry (just kidding)
I dig Natalie Portman and Clive Owen in the strip club scene in The Closer, as well as Natalie Portman and Zack Braff in Garden State.

reese witherspoon and ryan phillippee. cruel intentions. i always think it's neat to see how real life couples interact in movies. especially when they have to be mean to each other. you could tell ryan was getting really upset. i also think the two people that starred in the notebook had amazing chemistry.
