Get some activities for him. Get a exercise pin for him and leave him out for about 2 hours a night. Guineas like small rubber balls, or you can even get him an exercise ball. If worse comes to worse get another guinea, he'd love the company.

First of all, make sure you are not using a wheel or run around ball, guinea pigs arent like hamsters/gerbils/mice etc. They can break their backs very easily.
Also, dont use vitamin drops in water, if you are giving quality pellets- http://www.oxbowhay.com unlimited hay, and fresh veggies (in moderation) than there is no need to add anything to their water. Vitamin drops can cause a lot of problems in the long run.
Sorry, had to add that first because I have seen a lot of people telling other people to use balls/wheels and vitamin drops.
For her overweight problem, make sure you have a large cage (the requirements for 1-2 pigs is 28in x 56in) if you have a petstore cage, that is most likely the reason she is overweight. Check out guineapigcages.com for information on inexpensive but HUGE cages called C&C.
Another reason might be that she doesnt have anyone to play with, the best thing would be to adopt her a friend (of the same gender) so that they can play together when you are not home.. etc.. Adding another pig really doesnt increase the cost/work.

If the guinea pig has a large cage, you can put some rocks and bricks in the cage so he can climb on them and at the same time get some exercise will doing that. Take him to your room and let him run freely and use a treat attached to a string to make him walk after the treat for a while.

DON'T GET AN EXERCISE BALL OR WHEEL for the piggie, their anatomy can't handle that kind of activity. The webs below will give you information an piggie nutrition. If they don't have the proper room to move around all they can do is eat and gain weight. 4 square feet is need for one piggie. Once in a while a piggie will get pretty fat and stay that way.

Is it possible to have a larger cage On the outside? If so, the pig might be more motivated to get up and run around.

Are you sure he is really a he? One way to find out if he is a boy (if very young) is to run your finger over the skin above his peepee. If you feel a long, thin, narrow lump, it is his urethra na dhe is a boy. *hope that makes sense*
