It means that the person or entity has been wealthy for many generations. In your college example, it means that this college has had a large endowment and/or rich people attending for many, many years. This would mean that it's got 200 year old, beautiful buildings, ivy covered walls, classical amenities (big old fireplaces, crystal chandeliers, etc).

It's just a term to describe the "wealthy" that have been wealthy for a period of time, rather than someone like an entrepreneur who made money quickly.....
