Spin the tire with your hand while tightening the nut,when you start feeling resistance on the wheel, back off the nut until it becomes easy again.You do this to "seat" the bearing. After you loosen the nut about a 1/4 turn, check to see if there is any movement to the bearing by placing one hand at the top of the tire and one on the bottom and alternate pushing top then bottom.There should be a little bit of play, if there is too much tighten the nut until this play is acceptable.

It is a nut, not a bolt. With the wheel off the ground, tighten until there is resistance of rotation. Back off until the cotter pin can be engaged.
