I don't know. There are so many to choose from...
Some of the books of the Bible standout...Job comes to mind at the moment.
I can never decide if I like Persuasion or Pride and Prejudice better (Jane Austen).
Helle Haase's "In a Dark Wood Wandering" is haunting.
Connie Willis' "The Doomsday Book" is as well, but then her "To Say Nothing of the Dog" is totally hilarious (just like its inspiration "Three Men in a Boat...")
Nick Hornby's "About a Boy" is my favourite until I read his
"A Long Way Down".
And there are "Great Expectations", "The Old Man and the Sea", "Love in the Time of Cholora"...
Nope, I cannot name just one piece of literature that is THE best I ever read. Sorry. ; )

"The Scarlett Letter" by Nathaniel Hawthorne is my favorite.
I had to read it in college & I've read it at least another 5 or 6 times since. The movie (not the one with Demie Moore but the original) is filmed according to the book & is equally as good.
"Tiger Eyes" by Judy Blume is another favorite.
The girl in the story - I can relate to from my own personal experience. I read this one as a teenager for school. Even as an adult I've read it another 2 or 3 times along with my oldest daughter who has also read it & liked it.

To the Lighthouse by Virginia Woolf. Not so much for the plot as just the pure truth and power of her writing style.
(I so knew the first answer to this would be the bible! :P )
