well first of all unless there is a written policy stating that you can't smoke; your comapny is breaking the law. Also, locking oyu into an office is a safety hazard. If there was a fire or emergency you would not be able to get out. I would contact a lawyer. Your employers are clearly idiots.

You can quit smoking, delay the smoking until later, or, if it really bothers you, quit the job and get one that does not lock you in. Wherever it is that you do work probably has some sort of security issue. If not, there are limits to lock up situations, as in safety issues.

Yes you can quit smoking. That will improve your health anyway. However, my bigger concern is that you are "locked in". That is illegal ... a violation of fire codes at the very least. You should call the local fire department, anonymously if you're afraid you'll get into trouble (I'm paranoid so make sure you call from a number that is not your home so they can't check phone records) and report this.
