How would I set up a biology experiment to find out how pH affects the loss of pigment through the tonoplast and cell membrane?

Chlorophyll fluoresces when exposed to light, so there is an instrument called a flourometer that measures fluorescence of an aqueous sample. I assume that you are looking for chlorophyll that leaks out of the cell and into the water. If this is the case, then you could simply place equal volumes of an algae in several containers, each with different pH, let them grow for a number of hours, days, or some time sequence. Next, filter the algae out of the samples, and then do whatever is needed to analyze the samples in a flourometer.
