That's a perfectly legitimate question for a prospective employer to ask, and in my professional opinion, a good one.
Why? Because it tests the applicant's ability to tell me why I should hire that person as opposed to somebody else. True, you may have a degree just like my other applicants. And true, you might have similar work experience, but there's got to be something about YOU that separates YOU from the others.
That's what I want to hear! What that is all depends on your individual background, but you definitely need to take a cold hard look to find out what that would be.
For example, I'm one of the few in my department with a very broad work history. While many spent the majority of their time in one field, I bounced around a bit to find what I truly wanted to do, and this has given me a different way of looking at things. And a way that impressed this employer enough to hire me.

Depends on the positon. Usually it comes down to previous experience, knowledge, how much research you've done into the company and the positon you're applying to. Also how confidant you are and your general appearance ie. neat and tidy.
