If you are waiting for a download/upload to finish - make sure your laptop is on a well ventilated area, the laptop's AC adapter is connected, and your power settings are set to "always on" (so it doesn't hibernate). A few overnight should be fine but remember most laptops are usually designed for mobility (minimum cooling parts) and not intended for long continuous runs like desktops.

No. I leave mine on all the time. I would recommend putting it on standby though. You can do this by clicking Start > Shut Down, then clicking Standby, or just close the lid. Most notebooks automatically go to standby when you close the lid. This saves power and keeps the processor cooler.
