Pets or wild?
Adult rats are much bigger, of course, but a juvenile rat (3 weeks old) can look like an adult mouse.
Pet mice are mus musculus; pet rats are rattus norvegicus. The rat has a freckled tail, the mouse doesn't.
Most wild rats (in the city) are also rattus norvegicus, and you can tell them by the freckled tail. City mice are usually also mus musculus, and have a dark tail, often lighter colored underneath.
Some city rats are rattus rattus; these are a little smaller than norvegicus, but have a longer tail; and the tail is black, not freckled.

mouse is smaller and skinny tail, also small hands.
A rat has longer fingers and a fat tail,
Then there are the droppings,
a mouse looks like the size of the comma on your computer screen ( , ) and the rat dropping is the size of a piece of long grain rice. That is about 3 x the size of the mouse.

As other people have said, in general you can tell rats and mice apart by their size - rats are larger than mice. If you're dealing with young rats, though, it can be a little more difficult, since size won't tell you much there.
Young rats will have larger heads relative to their bodies than adult mice. Young rats' feet will also be larger relative to their bodies - adult mice will have smaller feet.

Mice are much smaller, and have smaller tails. Rats twice as big with thicker tails.
