If you mean engine oil, you can check it either, it is just easier to check it cold so you don't burn your fingers on a hot dipstick. But the car needs to be on level ground with the engine off.

If you mean transmission fluid, engine running and hot most cars with transmission in Park, that's if it is an automatic, sticks you can not easily check the fluid level.

If you mean coolant level, the overflow reservoir has cold and hot marks for fluid level, never try to open or check the fluid level at the radiator cap when the engine is hot.

you check it when its cold. Checking it right after you drive it will give you a misleading read because not all the oil has drained to the bottom. Best time to check would be in the morning because the car has been sitting and the engine is cold. It gives a more accurate reading.

Check the oil when the car is cold. If you are not having trouble, I would check every time I filled the tank.

Since the engine is running that means the oil is coursing through the engine. You will not get an accurate measurement because of this.

You check oil and transmission fluid not engine levels. Check oil while cool and transmission fluid while running and hot.
