Use a snake to determine the depth and approximate location, then work on that section of pipe. The snake will help determine where the first blockage is, and if it can't get through, other means are necessary.

Try a rooter...with a root cutter attachment. That should clear out the roots. The only way to avoid roots in the lines is to have the entire clay pipe sections replaced with PVC so there will be no seams. If you need to find the actual section, use the rooter until you find resistance...mark the rooter, withdraw the snake and measure it out on the surface to find where you met resistance.

We just run a sewer cable in and note where the roots are ,measure out that amount and you are close.
you can rent a camera and see what and where the problems are.

if you have a root problem there is only 1 way to fix it and it will either break your wallet or your back. Dig it up and replace the entire line from the house to the cities stack connection or your septic tank whichever you have.

a plumber often uses the flexible coil winder to clean your drains. this tool stops or slows down in this instance. unless you know a plumber its kinda doing it the hard way.

call a plumber with a power snake, well worth the price. then ask him what to treat with on weekly basis to eliminate their return.

You just gotta dig it up man, there is no easy way. Well unless you hire someone to do it. Its never where you think it should be.
