Well it depends what genre you're looking at.
Drama: My Sister's Keeper by Jodi Picoult
Mystery: A Thin Dark Line by Tami Hoag
Romance: Socrates in Love...I don't remember the author, sorry
Fantasy: The His Dark Materials Trilogy by Philip Pullman
Fiction: The Kite Runner by Khaled Hosseini
etc...Oh, and Flowers for Algernon was also very good, by Daniel Keyes ^^

The Hab Theory is my favourite book of all time, the author mixes fact with fiction to create a believable end of the world scenario.
I also have liked most of the books that have been mentioned so far.

There are lots of books actually. But from what I can recall right now is The Undomestic Goddness by Sophie Kinsella - it is a very funny book. There are other books too - Shopaholic series. Books by Lurlene McDaniel are touching and interestng.

The two I go back to over and over are Les Miserables by Hugo and To Kill A Mockingbird by Harper Lee.
But the truth is my decision of Best or Favourite changes daily.

I love many books for many different reasons, but if I have to choose the best, I'd pick the following:
"Becoming a Man" by Paul Monette
"Angels and Demons" by Dan Brown
"A Child called 'It'", "The Lost Boy", "A Man named Dave" by Dave Pelzer
"Going Solo" by Roald Dahl

I have 2 fav. books w/ totally different themes.
1.RUNNING WITH SCISSORS by Augusten Burroughs & 2.JACK THE RIPPER by Patricia Cornwell
Running.....Is a totally dysfunctional true story and Jack....is the true story(from Ms. Cornwells view) of Jack the Ripper and who it may actually have been. I will warn anyone that reads it tho, it has very graphic drawings.
Both books are very well written. Enjoy both!
