Disk Cleanup gets rid of unnessecary files on your harddrive, such as internet termporary files, recycle bin files, windows temporary files, old compressed files, offline files and so on, it gives you the choice as what you want to get rid of once you initiate the disk cleanup command, then the disk defragmenter is to tool that pretty much compacts your files together on your disk so that the disk can access your files easier on your disk drive, which will lead to your computer working a little faster or much faster depending on how badly your system if fragmented, when you system is fragmented its the same as you having an filing cabnet in your office and you having every file out of order and of course it will make it hard for you to find your files quickly, well thats the same with your hard drive, when your harddrive or your disk drive is fragmented it makes it hard for your system to locate files quickly which thast why it makes your system slower if you computer is fragmented badly, so thats what the defragmenter does it organized your files on your disk drive and makes it easier for your system to access them

To make your PC run faster you need to optimize the data being accessed on the disk surface. If that means deleting unnecessary stuff (Disk Cleanup) or re-organizing the data blocks that may have been moved around by other data blocks (Disk Defragmenter). As you can see they both go hand in hand. Because after you run your clean up there are gaps on the disk surface (only the computer can see that). These are known as "data fragments". So you "De-Fragment" those data blocks which reduces the need for searching the entire disk surface thus speeds up your PC.
FYI: Unix based filesystems, do not have a defragmenting tool. That is because they defragment on the fly! That is why Linux, for example is much more stable then windows. Umongst other things!

A process (run by a defragging program) whereby parts of data files on all segments of a computer hard disk are taken from their fragmented state (with parts of files spread all over the disk), and grouped together in complete-file segments. This makes it quicker for applications to find the files they need and frees up disk space, making the computer run more efficiently.

They don't make your computer significantly faster, but what disk cleanup does is get rid of all your cookies and temporary files. Defragment puts files back into their proper place

Defrag reorganizes your filesystem on the disk. As you save new files and delete older ones, the dik becomes crowded and large files are saved on two or more locations on the disk. Defrag joins every file and tidies up your physical disk usage.

disk cleanup removes cache files and such from IE, disk defrag resorts files so they are all together instead of bit and pieces all over the place.

The disco clean out deleates files from your computer to restore it to its factory settings (as it came the first time you took it out from its box) the Defragmentation process joins the info together in clusters (where you store your data) so that they can be accesed quickly and this process will not erase any data at all it just puts the pieces together (like a puzzle)
