Grow the mutant and some wild type bacteria with and without an inducer like IPTG or lactose and then do a beta-gal assay

LacIS mutants will have a much lower beta-gal acivity than wt under all conditions. By doing a beta-gal activity vs inducer concentration curve you can find out how much stronger is the effect of the mutant repressor.

LacIR will have the same amount of beta-gal activity in both cases and it will be approximately the same as that for wild type bacteria in the presence of inducer.
