Physically, yes, it is possible. The main reason it's not done is that the falls are a major tourist attraction, and if there were no falls, a lot of people would no longer travel to the area, and a lot of tourist dollars would be lost.
Also, half of the falls belong to the US, and half to Canada, so it would have to be agreed upon by both countries....

We already get a massive amount of electricity out of the falls. (See link below). 25% of New York State and the province of Ontario power requriements are met by hydro electric power (mostly from Niagara Falls).

There is no way the Falls could supply all of our electric needs. The fact is that there already is an electric power plant at Niagra Falls.
