I've played once at BGG.con 2006, and here are some strategies that I saw come up in the game play

1) I found the routes running along the left-hand of the board especially useful. I started the game with two routes along that way, and by mid-game had built a route from Kiel-Hamburg-Bremen- Muenster-Dortmund-Dusseldorf- Koeln-Frankfurt-Mainz- Mannheim-Stuttgart
This route especially proved to be especially great in the last third of the game, as I kept drawing short route tickets, and every time I did, I had already completed or was one step from completing at least one of the routes I drew. This not only netted me the points from completed route tickets, but it also handily won me the 10 bonus points for the most tickets completed.

2) Another player, early on in the game, executed a great route with one of his passengers. He had collected a couple of passenger tickets, and he was the first to run a passenger and picked up lots of the bonus chits. Since he was first, he got a lot of the top value ones, including the sweet 7 chit from Berlin. I believe he picked up some 30 points on that play alone. So remember, if other players are making connections and have built a network, all you need are passenger tickets to run your passenger over their rails to pick up the good chits.

3) For outright point gathering, there are the big-big routes -- a couple of sevens and sixes. Thought these don't have the same cache as completing ticket routes, they do stack up the points, and if you manage it right, you can dump all of your trains on the board while other players are left holding the bag on 1/3 to 1/2 of their trains (and they might not finish their routes either). Additionally, the new 4+ wild cards help this strategy significantly, as you can pick up a 4+ wild from the face-up cards just like a regular train car, rather than be limited as you would be for picking up a regular wild card (but the 4+ wild is only useful on routes 4 trains or longer... and there are a lot of those on this map). If you do opt for this route, it would be better to keep your short route tickets from the beginning. If you can finish them, that's great, but if you can't, then you want to keep the damage to your score at a minimum.

4) And, as with the other Ticket to Ride games, if you see a player needs to make a critical connection, take it before they can (Make them work around!) or if they are eyeing a particular route and are picking up corresponding color cards, then if you can deny them cards by taking them yourself, then their delay is your gain.
