I have several favorites....
a. the new Pride and Prejudice. They did an excellent job of staying true to the book! It is also an incredible romance story!
b. Just Like Heaven. Makes me cry every time I watch it.
c. Sound of Music. Julie Andrews. Nuff said.
d. Runaway Bride. I loveeee Julia Roberts in this!
e. ANYTHING Disney. =)

I can't answer this question, with more than 100 years of great cinema behind, starting From Murnau, Griffith, Ejsenstein and Bunuel... I can just say that I am almost sure that the most significant to cinema history and technique is Kitizen Kane... that doesn't mean it is the best one or the one I like most...

I have just seen John Tucker Must Die, and I think that it is hilarious. That is probably my favorite movie so far.
