I want to ask what is the main purpose of the second interview ? and what are the the expected questions

Generally the first interview is to see if you are qualified to do the job that you have applied for.

The second interview is to see if you are a good fit at the organization/dept that you will be working in.

The questions will be more detailed to the location that you will working at and the material that you will be covering.

You may have a panel interview with your future boss and two other representatives of management and they shoot questions at you. It might be just a one on one interview but possibly one with your future boss and maybe another collegue.

The best thing is know as much as you can about the job and the organization that you will working for, check the organization's website for information, know that profession, and know your skills (strengths & weaknesses), and how you can help that organization/dept.

They know you can do the job, this is about you proving you belong there.
