I'd say your brain & your eyes, but for software, it would be what you know best (be it photoshop, painter, illustrator, corelDraw!, flash, GIMP, Maya, 3DS Max, sketchup, or whatever).

Pick a raster program & a vector program (photoshop & illustrator are the standards) and have fun! :)
