Complicated answer...the item itself (shortcut icon) takes up no memory (RAM), but takes an extreemely smal amount of disk space. Its just a shortcut. You can have a million shortcuts on your desktop and it wont affect anything or slow you down.
Now, this shortcut may represent a running/active program which DOES take up memory. If you delete the shortcut from your desktop, the program is still running in memory. The application itself would need to be removed from in the control panel. This way, it removes the desktop shortcut icon (some uninstallers dont remove though), removes the application from the disk, and finally, stops the background tasks.
When you restart again, its not taking up memory, disk space, or CPU cycles.

Yes, they do on the Hard Disk but the space taken up is small. To find out the size, right click on a shortcut icon on your desktop and click on 'Properties', General.
