I think he knew all along but was heavily clouded by the Dark Side, whereas Luke knew he had Jedi potential therefore being able to realise it for himself making Vader less clouded.

i guess its because the emperor made him believe that padme is already dead which obviously meant the children must be dead as well. about the midiclorion levels perhaps as a child, leia wasn't given proper training so that the force might be strong in her. Remember in episode 1 when quin-gon jinn found anakin. He already thot that anakin's already too old for training. I mean look at luke, he started his training so late that he doesn't seem to have that much 'force' in him.

vader thought that padme died before giving birth so had no idea he had children until empire stikes back when he fights luke.
leia didn't know vader was her father when he boarded her ship to capture her so her thoughts couldn't give her away like luke's do in return of the jedi.
