You will just have to ask, and if they're not in the closet, they will tell you.
But if you are asking about yourself, check your attraction towards the same gender if it already has a long track record. Check if you have wondered for more than 10 times what your sexuality really is. If you have been attracted to the same 90% of the time, then you must be gay. If there have been both same and opposite sex attractions, then you're bi and so on.

Well you can't tell by looking or guessing. You can ask, but if they aren't out they won't tell you unless they trust you a great deal. If its someone you want to go out with, just ask them out -- if you yourself are gay and want to ask another boy out -- be a little careful, if he's a bigot it could go poorly, become his friend first and work up to it over time. If you are straight however, the girl will usually say no when you ask if she isn't interested.
If you are asking for yourself, if, when you aren't trying to block it out, you are sexually aroused by other boys, then you are probably at least bisexual. The first trick is to get past your fear, its just a learned reaction from adults anyway. Let yourself be attracted to whoever you are attracted to -- and soon enough you will know who that is. You can't change anyway, just make your life miserable for years.

If a person is having sex with a member of the same sex, he or she is possibly gay or bisexual.
If a person tells you he or she is gay or bisexual, he or she is likely gay or bisexual.
There is no other way to tell. There is no test, and "gaydar" is far from definitive.
