The key of a song is the chord, major or minor, that the song sounds like it is gravitating towards. Songs tend to sound like they want to resolve, or end on a certain chord, as a result of the harmonies, or chord progressions, that are being played (or implied). Lots of classical songs have the key of the song as part of their title (like... Sonata in E major, or Waltz in F minor).
Theres no hard and fast rule to determining the key of any song, although for most songs it is fairly obvious once you know what you're listening for. Many songs end with the chord that is the key of the song.
For example, consider the songs Happy Birthday, or Auld Lang Syne, .. Notice how the second to last note of each song really wants to resolve to the final note and ending chord? Sing them slowly if its not obvious. That final chord is the "key" of the song.
Sometimes a song will change keys one or more times during the course of a song. This is called "modulation".
A song can be "transposed" to different keys, making the song higher or lower in pitch throughout. Although a song might have a certain key that its usually played in, it certainly can be changed higher or lower. For example, a musician might transpose a song to a lower key if the song is too high for the vocalist to sing.

Although the key of a piece may be named in the title (e.g. Symphony in C), or inferred from the key signature, the establishment of key is brought about via functional harmony, a sequence of chords leading to one or more cadences. A key may be major or minor; music in the Dorian, Phrygian, and so on are usually considered to be in a mode rather than a key. When a particular key is not being described in the English language, different key naming systems may be used.
