Like Bush, JFK was a heirhead, an ignorant loose cannon who almost started World War III to flatter his spoiled ego. One goal the CIA and KGB had in common was to prevent nuclear war. So, together with the Mafia and other groups to present a confusing picture, the deal went this way:

The Russians would get rid of the Cuban missiles if the Americans got rid of the Turkish missiles. The Russians would get rid of Khrushchev, also a loose cannon, if the Americans got rid of Kennedy.

I think the Pro-Castro argument is rather obvious. The US government has attempted to assassinate Castro on at least 87 occassions, and failled, so why shouldn't Castro try the same.
The Anti-Castro affair is because during the Bay of Pigs fiasco, where the CIA tried to use Cubans to overthrow Castro, JFK refused the US military to use air support (which would have been an open hostility, and as such a declaration of war by the US) to help the disidents.
The Mafia were angry that Robert Kennedy had directed policing to break the power of organised crime, and as such any member of the government and Kennedy clan was regarded as an enemy.
The CIA, as I've mentioned, had the same argument as the Anti-Castro lobby, because it was the CIA who stirred up the Bay of Pigs fiasco.
There is also another reason which was hidden from the US people why either the CIA or Mafia may have been paid off to kill Kennedy. Kennedy won the election because he was seen as a young, fit and healthy charismatic Christian with a nice family. In the run up to the elections after his death, Kennedy could not have kept this false persona up. He was racked by ill-health, most notably back problems and syphilis. He was on an extreme dosage of pain killers, and could not get out of a chair without assistance. His affairs with prostitutes and anything in a skirt (makes Bill clinton look saintly) were becoming too well-known, certainly amongst his opponents, the media and the CIA. He was still drinking heavily, which with his medication was making it impossible for him to hold a proper discussion, never mind a face-to-face political debate. Rather than have his party exposed, and probably a heavy loss in the elections; it MAY have been a better option to simply have this sick animal 'put down' for his own (and his partys) good, hence making him into the martyr and saint that most Americans think of him still today. And, of course, winning the sympathy votes at election. The fact that his brain was misplaced after the autopsy, when it would have clearly shown the extent of his alcoholism and syphilis, has added to this speculation, especially as his medical records were not releasd till 30 years after his death, and detail his illnesses in graphic contrast to the media myth.
