You need a good drawing program like illustrator or photoshop and an animation program if you want your anime to move. Anime is a specific technique that exaggerates eye size so they take up most of the head size, use a barely perceptible nose and mouth.
How to create virtual beauties is a good book but you have to have the software to use it.
