A circular route:
Get the tube to Trafalgar Square.
Walk down Whitehall past Horseguards and Downing Street.
Visit Westminster Abbey and Houses of Parliament.
Then get a boat from by Westminster Bridge along the Thames to The Tower of London.
Walk to StPauls via The Monument.
Get the tube to Green Park.
Have a look at Buckingham Palace and take The Mall back to Trafalgar Square.
If you then have any energy left you can do some shopping in Oxford Street.

I would go to Westminster Abby and the Houses of Parliament, then cross the river and go on the London Eye ferrous wheel.
Take the underground tube system and also check out Oxford Street for some shopping or Knightsbridge for Harrods department store. If you have time, go see the Tower of London and then from there you can take a short river cruise.

Actually it is also a great idea to get on to the London sightseeing bus. I think you can buy a ticket on line. The bus takes you all around the city, with live commentary, and you can get on and off as often as you like during the whole day. That way you get a great overview of the city and things you may enjoy doing.

Being in London is an experience in itself. I love the city and you need to absorb it, not just go place to place. Riding the underground is a great experience (its also the best subway system in the world, so easy) and definitley go to parliment and westminster, Tower of London, find a nice pub and enjoy a real ale (its pumped up not shot out of the spout) and some traditional pub food. If you have more time there are 2 great war museums one is called london (or england) at war and the second is Churchills bunker, both are great but take a good while to get through. Enjoy your day and try to get back for an extended stay.

Go to the Tower of London, Saint Paul's Cathedral, and the London Dungeon (it's a small, semi-campy museum about the bloody history of London). I would also see a play on West End. And, eat lots of fish and chips. I love fish and chips. :-)
