It depends on the level/quality of the relationship. If you trust her then let it go. If you don't' but be prepared for what you may find.
Usually when you dig/look for dirt you find it but also get dirt on you (hurt) in the process.
Calling will not necessarily give the answer. If it is a man it could be Innocent contact for a job, relative etc. If he is seeing her there is a good chance he will deny it. So what would you have gained?
I would save it and wait till I got more concrete evidence to confront her with.
