If i took a cup and slid it down a ramp and did this 5 times....but added 100ml of water to this cup...How is the kinetic energy of the rolling bottle affect the amount of work done on the cup?

the kinetic energy really has nothing to do with it. It is the force of rolling down the incline that does work. The kinetic energy will increase (as does the work) as water is added to the system, however kinetic energy really doesn't do anything in the lines of work.
