The best way is to go to RUN, then type MSCONFIG and hit enter. Uncheck everything under the STARTUP tab and click APPLY. Restart your computer, and it goes much quicker without a bunch of crappy unnecessary programs running. Also, under the control panel, defragment your hard drive.

Memory is freed every time you close a program or restart Windows. You can also add more ram (physically) to combat a system shortage. To free up Hard Drive space, uninstall unused programs, run a Window's drive sweeper to delete garbage files, or buy a bigger Hard Drive if your total files eat into your swapspace.

Most of the time PC's that get "slow" over a period of time are plagued with spyware/adware. Get Spybot and check to make sure no malicious software is present.

Try to defragment your hard disk, it often helps much.
