It would depend whether you return those feelings or not. I know two people, a man and a woman, who've been very good friends for many years. She's deeply in love with him, has loved him all those years and suffers horribly when he pays attention to any other woman. He appears unaware that she has these feelings for him but it's crystal clear that he would never feel the same about her. He likes younger women who are very pretty and his friend is approaching her 50s, makes no effort whatsoever with her appearance and looks as if she's never heard of sex. If she told him how she felt, I believe it could mean the end of their friendship. At best it would radically change the nature of that friendship. Whether it would be best for her to face the truth, I don't know.
Only you know how you and your friend feel about each other and ultimately only you can decide.

If my best friend was a guy:
I'd tell him, cool, but I'm not into masculinity. We can be friends though, but repect my heterosexuality. Then I'd ask, why? Are you just confused or for real? If for real, let him know he may get hurt if he dwells on that feeling for me when I find the right girl. He should go else where to find the right guy.
If my best friend was a girl:
We'd get in on. In fact, my best friend is my wife :)
