It depends on what motherboard you have and what it supports. Number one, if it doesn't support Crossfire AND you want to have two video cards installed then yes, you will need to upgrade it.
If you only want to install one video card then it doesn't matter... it doesn't matter if you have a PCIe slot. If you don't have a PCIe slot then you will have to upgrade your motherboard. Crossfire cards only come in PCIe versions so if you don't have a PCIe slot then it won't matter either way, you won't be able to use it.
Likewise, if all you have is an AGP slot then you won't be able to use Crossfire or SLi cards... but there are plenty of current generation cards available in AGP variants.

no, crossfire just means u can run 2 different ati cards on the 1 board, compared to SLI which need to be the same cards.
