It is an informal expression used in the same way as 'wake up call'. In other words, it means to knock someone's door to wake that person up! So, do you want me to knock you up because you need to be in time for the examination which starts at 8.30 a.m.?

I`m Canadian and even I know this one! All of the above answers relate to the American meaning. In the UK it means to wake you up ie. I`ll knock you up in the morning.
