Perhaps get in touch with a temp agency in the new city. At my company we will do a "temp-to-hire" a lot of times with assistants so you may get a chance to do that.

Look on Monster.com for Assistant Jobs in Chicago. Send them a coverletter and resume. Tell them you want to relocate to Chicago and you saw their job posting. Tell them the dates you plan to be in Chicago for interviews. I did that when I relcoated out of state and got an offer on my first interview. Good luck.
