No, you're mixing up simple slope, change in y over change in x, with the process of finding the entire equation of the line through those points, for which finding slope is just the first step.
So with (4,6) and (2,-1), subtract y's, -1-6=-7, and subtract x's, 2-4=-2, and get slope -7/-2 = 7/2.
To get the equation of the line, THEN you solve
y = (7/2)x + b with (4,6) plugged in for x and y.
6 = (7/2)(4) + b
6 = 14 + b
-8 = b
So y = (7/2)x - 8
