I'm guessing gram stain is one of them....

try API20E or any other commercial available kit. or conventional sugar test recommended by CDC.

Since my wife is a clinical microbiologist her answer is you must look at the sample you received and the clinical data before deciding what your next step would be. Her lab does about 1,000 samples a week.

Certainly a gram stain may be one of them but you did not give us enough information to answer your question thoroughly.
