'One-night stand' refers to a sexual encounter that lasts no longer than a night. Generally, it is with someone you do not know beforehand, but always means that whatever connection you had will end at the end of the night, and you will not be with the person again.
So basically, you meet a stranger, have sex, and never see them again- that's a one-night stand.
