The cheapest transport for one person is a Skybus to Southern Cross Station and getting a train or a tram to the whatever hotel you are staying at, it will cost about $15 a person but is inconvenient if you are not in the CBD and carrying lots of luggage. A taxi is likely to be at least $50 from the airport.

If your talking about the bayview hotel in south melbourne, i'd probably take the shuttle, just because the taxi's can be less than perfect. At least this way you know you will get there.
