I would advice you not to travel by road. The road from Mumbai to Nasik is very bad that too after this monsson it has got crores of potholes and at some places the roads are so narrow that if a heavy truck passes then you'll end up in a huge traficc jam. Any way if u still decide to go you can take a the western express high way till nasik. It will take minimum of 4.5 hrs to reach Nasik from there you'll take another road to shirdi which is approximately 100 kms and will take 2 hrs.
I would suggest you to go by train.
2859 Mumbai CST-Howrah Geetanjali Superfast Express wtakes 3.5 hrs to NAsik Road and from there you can take private taxis.

Its better to take a neeta volvo from mumbai to shirdi at 9.30 am. You check for the return trip time in the same bus.

I think you can take a bus Usually by car it takes 5 -6 hrs.
By train you can go to manmad from there take a taxi it takes 45-1 hr from there or go to Kopergaon from it is almost 20-30 minutes.train journey is also 5-6 hrs To directly go to shridi take bus or car.I think roads are good for transport.
