RNAi is the natural process whereby an organism can suppress its own mRNA transcriptions' translation by forcing anti-sense RNA onto it, blocking the coding mRNA. It is also done by researchers to purposefully "knockdown" a gene to see what happens next.

Generally, for RNAi, the sample is treated with the RNAi antisense sequence, and then the presence of a "downstream" protein is analyzed.

The sample to be treated is often a flask-based cell cuture. That's the only one I've seen anyways.

That can be done by Western blotting (to get a "yes/no" answer on the expression of a protein), by ELISA (a method with great precision, though fumbly accuracy).
