cancer is the big one, cancer cells are cells that just don't die, so they grow and reproduce take up valuable nutrients from healthy cells, crowd out healthy cells until the healthy cells die, and finally take over your body until it's systems shut down.

Sickle Cell? Sickle cell is caused by a mutant gene for the alpha/beta subunits of hemoglobin.
Cancers come about due to mutations in cell cycle regulation proteins. p53, cyclin dependant kinases, etc.
