Cape Town, most definitely, or Durban.
I myself like the cape area best, there are many nice towns between Cape Town and Port Elizabeth, the people are generally nice, the food is cheap and lovely, so is the wine.
I recently spent 4 weeks in Cape Town working in an orphanage, and I enjoyed it a lot, in spite of the dreadful poverty which is still widespread, especially among black people.
As a tourist you can just stay out of these areas, and you will find Cape Town a most enjoyable place. Don't forget to visit the aquarium, it's spectacular.

Hmm, asking for one place to visit is a tough one. Ok, so I'm gonna tell you about Joburg:
Firstly don't listen to what other people say about not visiting Joburg. I personally live in Joburg and I am not planning on moving anytime soon. What does joburg offer? Well, there is Gold Reef City amusement park which is great. It also contains other things such as a circus, various shows, a gold pour where they show you how gold is made, a 4-D theater, hotels, trips down into a mine because the park is built on an old gold mine site. Then from there you could go on a tour of Soweto which is just around the corner. There are also the black townships which usually makes good tourist sightings. We also have a string of night clubs which are great and luxurious hotels in the big business district area. There is the famous Joburg zoo which is a must visit too! The crime is not as bad as people make it out to be and the scenery is great. You may as well come to Joburg for your visit! You could even move around South Africa as you choose for low costs, especially if you are coming with foreign currency! Hope you have fun!
