The advantages are that they can be very precise. In other words, specific access can be granted on the basis of palm print, thumb print, retinal scan or other biologically unique features.
The drawbacks are :
1) Cost
2) Operational...in other words, if i have a communicable disease and I put my palm on the same pad you do....
3) False positive results etc...
It is hard to calibrate the machines. Either they require the match to be too precise and dont let anyone have access, or they are too loose and let anyone in, hard to strike a balance.
