Currently, my desktop PC, when in standby mode, can only be woken up by hitting the computer's power button. Is there a way to configure the computer to wake up out of standby by just pushing one keyboard button or by clicking the mouse?

Thanks.

Hitting buttons does NOTHING. That's the point... I'm trying to get it to wake up by pressing a button. How do I allow this setting?

go to control panel.
there go to keyboard(double click the keyboard icon in control panel), a window will appear.
click on hardware, then on properties, a new windows will appear
There click on power management and then select the option saying "allow this device to..."
then press Ok.
hope this will work.
this does work in XP

I don't know for sure about stand-by mode, but I can turn on my computer by pressing a series of keys at home. I enabled this through my motherboard BIOS. You can try it and see if it will work for stand by mode the same as for a cold power-up. To get to my BIOS I hit the delete key while my computer is booting up. It's different for each motherboard, so yours may be the delete key or it may be another such as F1 or F2 (those are the 3 most common.) I would test it myself, but I'm not at home now.

Standby is a state in which your monitor and hard disks turn off, so that your computer uses less power. When you want to use the computer again, it comes out of standby quickly, and your desktop is restored exactly as you left it. Use standby to save power when you will be away from the computer for a short time while working. Because Standby does not save your desktop state to disk, a power failure while on Standby can cause you to lose unsaved information.

Open Power Options in Control Panel.
In Power Schemes, click the down arrow, and then select a power scheme. The time settings for the power scheme are displayed in System standby, Turn off monitor, and Turn off hard disks.
To turn off your monitor before your computer goes on standby, select a time in Turn off monitor.
To turn off your hard disk before your computer goes on standby, select a time in Turn off hard disks.

you should be able to set one key options
